چکیده

CD40 signaling in B cells and dendritic cells (DCs) is critical for the development of humoral and cell-mediated immunity, respectively. Nuclear factor k B (NFk B)–inducing kinase (NIK) has been implicated as a central transducing kinase in CD40-dependent activation. Here, we show that although NIK is essential for B cell activation, it is dispensable for activation of DCs. Such data provide compelling evidence that different intermediary kinases are used by different cellular lineages to trigger NFk B activation via CD40.

Lineage-Restricted Function of Nuclear Factor κB–Inducing Kinase (Nik) in Transducing Signals via Cd40

CD40 signaling in B cells and dendritic cells (DCs) is critical for the development of humoral and cell-mediated immunity, respectively. Nuclear factor kappaB (NF-kappaB)-inducing kinase (NIK) has been implicated as a central transducing kinase in CD40-dependent activation. Here, we show that although NIK is essential for B cell activation, it is dispensable for activation of DCs. Such data pro...

Negative regulation of the nuclear factor kappa B-inducing kinase by a cis-acting domain.

Nuclear factor kappaB (NF-kappaB)-inducing kinase (NIK) participates in the activation of NF-kappaB, a family of eukaryotic transcription factors that mediate cell growth and transformation. NIK activates the IkappaB kinase both in vivo and in vitro, although how the activity of NIK is regulated has remained unclear. Here we show that the N-terminal region of NIK contains a negative-regulatory ...

A Complex Relationship between TRAF3 and Non-Canonical NF-κB2 Activation in B Lymphocytes

The adaptor protein TRAF3 restrains B cell activating factor receptor (BAFFR) and CD40-mediated activation of the NF-κB2 pathway in B cells. Mice lacking TRAF3 specifically in B cells revealed the critical role of TRAF3 in restraining homeostatic B cell survival. Furthermore, loss-of-function mutations of the traf3 gene have been associated with human B cell malignancies, especially multiple my...

TNAP , a Novel Repressor of NF - B - inducing Kinase , Suppresses NF - B Activation

NFB-inducing kinase (NIK) has been implicated as an essential component of NFB activation. However, the regulatory mechanism of NIK signaling remains elusive. We have identified a novel NIK interacting protein, TNAP (for TRAFs and NIK-associated protein). In mammalian cells, TNAP physically interacts with NIK, TRAF2, and TRAF3 but not IKK1 or IKK2. TNAP specifically inhibits NFB activation indu...

Developmental expression of tyrosine kinase b in rat vestibular nuclear neurons responding to horizontal and vertical linear accelerations

Brain-derived neurotrophic factor (BDNF) is known to be crucial for the development of peripheral vestibular neurons. However, the maturation profile of the BDNF signal transducing receptor, tyrosine kinase B (TrkB) in functionally activated otolith-related vestibular nuclear neurons of postnatal rats remains unexplored. In the present study, conscious Sprague-Dawley rats (P4 to adult) were sub...
